Monocytes: players of rheumatoid arthritis and therapeutic targets
Our group is interested in monocytes differentiation into osteoclasts (OCs), multinucleated giant cells specialized in physiological and pathological bone resorption. The OCs activity is increased along rheumatoid arthritis (RA) giving birth to bone erosion and destruction.
The phenotype of monocytes and their modification by therapeutics along RA are not known. We study the capacity of RA treatments (anti-TNF, anti-JAK2) to modulate the polarization of monocytes. The transmembrane TNF-alpha (tmTNF) can induce a reverse signaling. We have demonstrated that anti-TNF are able to induce by reverse signaling an increase in CD36 expression and are able to inhibit the TLR4 mediated-ROS production in monocytes. Our projects are aiming at understanding the mechanisms involved and the consequences on RA treatment.
