PROJECT : INFLAMMATORY BOWEL DISEASES AND MACROPHAGES POLARIZATION
We are studying the role of macrophages in inflammatory bowel diseases and particularly their polarization and implication of C-type lectin receptors in the course of intestinal chronic inflammation. We identify signaling pathways and molecular mechanisms that control the M1/M2 polarization of macrophages.
ANIMAL AND CELLULAR MODELS
Primary cell culture :
- Human monocytes-derived macrophages
- Murine peritoneal macrophage
- Murine bone marrow derived macrophages
- myeloid cells specific deficient (LRH-1, PPAR-gamma, Dectin-1, Mannose receptor)
- Knock-out mice (SignR1, SignR3, STAT6, NRF2, IL-13Rα, 12/15-Lox, 5-Lox)
- Experimental murine model of inflammatory bowel disease and colorectal cance
- Infections : fungal and parasitic
TECHNIQUES AND METHODS
- Phenotypic and functional characterization of Macrophages:
- Flow cytometry
- RT-qPCR
- Isolation of colonic macrophages
- western-blot, ELISA, EIA lipid quantification
- ROS and NO production
- MPO
- microbicidal activities…..
